說明：CellG5檢測試劑盒用于檢測纖維素內(nèi)切酶（1,4 -β-葡聚糖內(nèi)切酶）包含兩種組分：1) 4,6-O-(3-Ketobutylidene)-4-nitrophenyl-β-D-cellopentaoside (BPNPG5) 和 2)熱穩(wěn)定的β-葡萄糖苷酶。 酮阻斷基團(tuán)可阻止β-葡萄糖苷酶對BPNPG5的水解。通過纖維素內(nèi)切酶的培養(yǎng)可生成非阻斷的顯色寡糖，被β-葡萄糖苷酶迅速水解。產(chǎn)生4-硝基*的速率與β-葡萄糖苷酶水解BPNPG5直接相關(guān)。加入Tris緩沖液(pH 9.0)后可終止反應(yīng)，并形成*顯色產(chǎn)物。
CellG5檢測法代表了代表了纖維素酶測量方法的巨大進(jìn)步。傳統(tǒng)的纖維素酶檢測依賴于酶底物，例如羧甲基纖維素，微晶纖維素，纖維低聚糖，CMC濾紙，染色多糖，包括CMC剛果紅或纖維素天青。
